The quartet of Alexander Beaton, Luke Belton, Miles Munro and Tom Hollingsworth finished over a second ahead of strong Jersey and Cayman Islands teams in the 4x50m medley relay, clocking 1-42.53.
That was one of eight medals for Guernsey at the National Sports Centre's pool.
The solitary silver came from Games debutant Courtney Butcher with an impressive performance in the women's 200m freestyle, clocking 2-03.56 behind established Jersey star Emily Bashforth.
Later Butcher joined up with Annalise Munro, Vicki Parfit and Leah Winberg to claim bronze in the women's 4x50m freestyle relay.
Two of the successful men's quartet also tasted individual success as Belton took bronze in the 100m breaststroke (1-05.26) and Beaton did likewise in the 50m backstroke (26.87).
There were also bronzes for Ollie Nightingale (men's 1,500m freestyle, 15-59.38), Ben Lowndes (200m butterfly, 2-05.52) and Jeremy Osborne (200m individual medley, 2-07.26).
